Best Setup for Good TVC/Broadcast at DVinfo.net

I have a deal in the works to make 'Build A Spot' TVCs (that's what they call it) for several political candidates around the country. I will mainly use AE CS5 along with Photoshop and Premiere. I have a very fast HP Z800, Eizo CG243W and a Panasonic BT-LH1710w, and I am planning on getting a Kona LHi to use with the Panny.

Many of these commercials will have a maximum 2 day deadline and they will be uploaded to an online distributor who sends the spots to the various stations.

So, can anyone recommend a good setup/workflow for quick turnaround and accurate broadcast legal colors, and any other pertinent info?
